Can I do NIOS and Diploma together?

Yes, you can do that. Each student is free to do one regular course and one correspondence course. So you can do a diploma course and class 12 NIOS simultaneously.

Is it possible to do Diploma and 12th together?

No, you cannot pursue both class 12 and Diploma simultaneously. As both are professional regular courses, one cannot pursue both the courses at a time.
